Mcm2-7 complexes were released from the beads by EcoRI digestion&#xd;
as previously described (Remus et al, 2009) and applied directly&#xd;
onto carbon-coated copper grids for single particle EM analysis after&#xd;
negative staining. Fig 1D shows that Mcm2-7 double hexamers were&#xd;
readily observed in both the treated and untreated samples and,&#xd;
more significantly, Mcm2-7 single hexamers were not found in&#xd;
either sample. This indicates that DDK phosphorylation was not&#xd;
sufficient to cause efficient dissociation of the Mcm2-7 double&#xd;
